Mapping of the 5-methyltryptophan resistance locus in Bacillus subtilis.

The 5-methyltryptophan resistance locus (mtr) in Bacillus subtilis, which leads to constitutive production of the tryptophan enzymes, has been mapped on the chromosome. The order of loci is ser-1-mtr-aroF-aroB-trp-hisB.

Characterization of the Bacillus subtilis tryptophan promoter region.

The nucleotide sequence of the control region of the trp operon of Bacillus subtilis has been determined. The region was shown to contain the trp promoter by deletion analysis and by determination of the transcription start site. The trp promoter shows similarity to the consensus sequence for Escherichia coli and B. subtilis promoters. The presence of the trp control region on a high-copy-numbe...

Control of tryptophan biosynthesis by the methyltryptophan resistance gene in Bacillus subtilis.

5-Methyltryptophan-resistant mutants derived from Bacillus subtilis strain 168 synthesize all of the tryptophan biosynthetic enzymes constitutively and excrete tryptophan. These mutants can be divided into three classes: class 1, low enzyme level and low rate of tryptophan excretion; class 2, high enzyme level and intermediate rate of tryptophan excretion; and class 3, high enzyme level and hig...

Metabolic interlock. The influence of histidine on tryptophan biosynthesis in Bacillus subtilis.

A regulatory relationship in Bacillus subtilis between histidine and the synthesis of tryptophan exemplifies “metabolic interlock,” a phenomenon of regulatory interactions exerted between biochemical pathways. The enzymatic basis for the histidine-tryptophan relationship was examined. Histidine activates anthranilate synthetase by increasing the apparent value of VmSx. Hence, histidine antagoni...
